I have made an .htaccess to my root directory for creating a subdomain level, assume it is sub.domain.ex that redirect to domain.ex/deb/
RewriteEngine On
RewriteBase /
RewriteCond %{HTTP_HOST} sub.domain.ex
RewriteCond %{REQUEST_URI} !deb/
RewriteRule ^(.*)$ /deb/$1 [L]
and this works well.
Now, I go to /deb/ and create another .htaccess with the following
RewriteEngine on
RewriteRule ^([^/]+)/ /deb.php?app=$1 [NC]
the deb.php is a file that prints the argument "app".
It works but only if i call http://sub.domain.ex/something/ (note the slash at the end). It only works with a final slash, if I remove it, it doesn't and I want it to work without final slash.
So I change the rule into ^([^/]+) but now I have 500 Apache internal error.
The regex coaches are by my side with the selection, maybe I'm missing something.

I'm runinng mad. Mybe is wrong to put one .htaccess in the root for creating se 3th sublevel doman and the .htacces in the other directory? Because I'm ttrying some trick.
I use this rule
RewriteEngine on
RewriteRule ^([^/]+)/ deb.php?app=$1 [NC]
in the .htacces of the /deb directory and made a print_r($_GET); and called index.php. So the redirectory doesn't work at all if it forward me on the index.php of the cydia sublvel and doesn't take the /deb/deb.php!!!

my dir structure is this:
/htdocs/ -> the root of the main domain level like www.example.com
---index.php -> home file of www.example.com
/htdocs/deb -> the root directory of the 3th sublevel domain (subdomain.example.com ->

---deb.php
So the .htaccess for the 3th level domain is the in /htdocs./htaccess and described as before.
The other .htaccess for "beautify" the link is in the /htdocs/deb/.htaccess. I want that when you go to subdomain.domain.com/someText it transform to deb.php?app=someText
Now i tryed go to subdomain.domain.com/deb.php....WTF!? the deb.php is in /htdocs/deb/deb.php

Edit: I updated the second file's contents to reflect your additional requests. You should remove the /? if you don't want them to be able to go to sub.domain.ex/something/ -> deb.php?app=something.
I could be wrong, but in the second .htaccess you should check that the url wasn't yet rewrited with "/deb.php"
So something like
RewriteCond %{REQUEST_URI} !deb\.php
RewriteRule ^([^/]+) /deb.php?app=$1 [NC]
You rewrite rule should be
RewriteRule ^(.*)$ deb.php?app=$1 [L,QSA]
That's a common pattern used in CMSs like Drupal
I think there is some kind of rescrctions to my host. It's impossible. I tried all kind of goo (for a regexcoach) for matching the correct group and there is nothing else the 500 error apache.
The only way to get this to work is use tu parameters
with this ^app/([^/]+) /deb.php?app=$1 and calling with sub.domain.com/app/nameTest and this is working with or without the end backslash.
Of you get some advice for getting rid of this, please let me know.

What I need to do is to redirect the files respectively to:
temp-file-1.php
temp-file-2.php
temp-file-3.php
...
temp-file-255.php
I know how to rewrite it one by one, but that is not as solution.
Can you help to rewrite these type of links in wildcard rule/cond?

Assuming keyword file is present in all the links, you can use this single generic redirect rule in your site root .htaccess to redirect all links:
RewriteEngine On
RewriteRule ^(temp)(file-.+\.php)$ /$1-$2 [L,NC,R=301]
(temp.+) is 1st group that matches starting temp
file-\d+\.php is 2nd captured group that starts with file and ends with .php
Try it like this, when working correctly you can change R=302 to R=301.

I have a multilingual wordpress website and want to redirect website of given region to given language,
xyz.de --> xyz.de/de/
xyz.co.uk --> xyz.co.uk/en/
direct access to xyz.de/de and xyz.co.uk/en are working properly. So there is no problem on wordpress side.
Now, I am trying to change the htaccess file of xyz.de and xyz.co.uk so that they redirect the website.
Considering xyz.co.uk
I want to add a RewriteCond such that whenever there is no /en trailing after xyz.co.uk it will automatically add /en.
For example xyz.co.uk/<trailing address> results in xyz.co.uk/en/<trailing address>
So far I have the following code, which somehow doesn't seem to work,

RewriteRule ^(.*)$ http://xyz.co.uk/en/$1 [L]
The negation of /en is not working! I have also tried

Could someone tell me where I am going wrong? seems like I have gone wrong in writing RegEx and suggest if there is better way to achieve the same, that does not affect the SEO across different domains.
Use THE_REQUEST variable instead of REQUEST_URI:
RewriteCond %{HTTP_HOST} \.co\.uk$ [NC]
RewriteCond %{THE_REQUEST} !/en/ [NC]

Make sure to keep this rule as your very first rule in .htaccess.
Change it to R=301 once you've tested.
I am using below code on my .htaccess file

the goal is to redirect a clean URL like below

The problem with your current rule is that you are rewriting unconditionally. Any URL that contains a single slash will get rewritten. I imagine that some of your (static) photo URLs match this pattern.
Common practise is to only rewrite the URL if it doesn't match an existing file (or directory):
RewriteCond %{REQUEST_FILENAME} !-f
RewriteCond %{REQUEST_FILENAME} !-d
RewriteRule ^([^/]+)/([^/]+)$ /view_basket.php?order_id=$1&pin=$2 [L]
This makes sure the request is only rewritten for non-existing files (not a file or a directory). I've also made the pattern a little more restrictive so there must be 1 or more chars before and after the slash (+), instead of 0 or more (*).

I want to redirect domain1.com to domain2.com in all cases except one particular case:
domain1.com/subfolder/index.php
I want this domain1.com/subfolder/index.php to be intact and not get redirected to domain2.com because I have hundreds of users already bookmarked this page.
But anything and everything besides that domain1.com/subfolder/index.php, I want domain1.com to be redirected to domain2.com

Use just [L] if you want the redirection to be transparent i.e. without letting your visitors know.
Use !^/subfolder/.*$ if you want to stop redirection for the complete folder as well as its contents.
You can use RewriteCond to check to see the requested uri is anything but the one you want to redirect, capture the desired path and then redirect to the second domain.
RewriteCond %{REQUEST_URI} !^/subfolder/index.php$
RewriteRule (.*) http://domain2.com/$1
I'm diving into the uses of htaccess/mod rewrite for the first time and I'm having a little bit of trouble with a redirect/mask.
I have a test directory in my root folder called modrw, in that folder is a index.php file with a nice and simple:
<?php echo $_GET['name']; ?>
In the browser if I type www.domain.com/modrw/{word}/ then the word is echoed on the page, which is what I want.
If I type www.domain.com/modrw/name={word} then I am redirected to www.domain.com/modrw/{word}/ and the word is also echoed as I intended.
However, if I direct the browser to the URL www.domain.com/modrw/?name={word}/ the word is echoed but I am not redirected to www.domain.com/modrw/{word}/ like I hoped.
How is the ? causing troubles? In the RewriteRule code below the ? is included, I've tried it a couple different ways but can't get it working.
Here is my htaccess file:

Remember that RewriteRuleonly matches REQUEST_URI. To match query string you must use RewriteCond with variable %{QUERY_STRING}. For ex. in your case:
